چکیده

Millipore MultiScreenTM filter plates are used for elution of dried blood spot samples in neonatal screening assays. A significant concern with the use of these plates is their cost. In this study we have evaluated the practice of reusing the filter plates in the Bio-Rad PKU and total galactose neonatal assays. All wells of the test plates used in the recovery study above were washed for a fifth time and then filled with DBS using the zero standard. An unused plate filled with zero standard DBS was used as a control. DBS were eluted and tested according to assay protocols. Results of the fifth run with zero standard on the used plates were compared to zero standard eluted on the unused plate. MATERIALS AND METHODS RESULTS Cleaning procedure Physical damage All dried blood spots (DBS) were removed from the wells by shaking or tapping the plate over an appropriate biohazard trash bin. The plates were rinsed under running tap water, ensuring the plates were filled and emptied 3 times. The plates were soaked for four hours by immersing in distilled or deionized water, ensuring that all wells were filled with water. Excess water was shaken out, blotted on paper towel, and dried in a 37 C incubator overnight.

Using the fluorescence spot test for neonatal screening of G6PD deficiency.

To establish the neonatal screening method of glucose-6 phosphate dehydrogenase (G6PD) deficiency, G6PD activity was measured using the fluorescence spot test (FST) using dried blood samples on filter paper. The G6PD/6PGD rate test of venous blood samples was further performed for confirmation. The positive G6PD deficiency rate was 4.2% and its detection rates were 3.7% for all neonates and 5.2...

Rapid DNA Extraction from Dried Blood Spots on Filter Paper: Potential Applications in Biobanking

OBJECTIVES Dried blood spot (DBS) technology is a microsampling alternative to traditional plasma or serum sampling for pharmaco- or toxicokinetic evaluation. DBS technology has been applied to diagnostic screening in drug discovery, nonclinical, and clinical settings. We have developed an improved elution protocol involving boiling of blood spots dried on Whatman filter paper. METHODS The pu...

Use of a dried blood spot in immunoreactive-trypsin assay for detection of cystic fibrosis in infants.

We assayed more than 5000 blood spots dried on filter paper and approximately 1000 serum samples for immunoreactive trypsin, with commercial reagents (Behring and Sorin). The assay procedures were modified so that newborn screening is technically feasible. Both kits are satisfactory for serum assay, but the Sorin materials are better adapted for blood spot analysis. Immunoreactive trypsin in bl...
